DESCRIPTION:Cancer cells display heterogeneous and dynamic states in gliob
 lastoma\, but how these malignant states arise and whether they follow a t
 ractable cellular trajectory across tumours is poorly understood. Here\, w
 e generate a deep single cell and spatial multi-region atlas of glioblasto
 ma that integrates transcriptomic\, epigenomic and genomic analysis to com
 prehensively characterise their tumour heterogeneity. We describe spatiall
 y-patterned transitions of malignant cells from dev-like towards glial inj
 ury response- and hypoxia-defined states during tumour expansion. This mal
 ignant cell trajectory dominates glioblastoma\, manifesting across tumours
  and genetically distinct subclonal lineages that are finely spatially int
 ermixed within tumours. Moreover\, this trajectory unfolds across speciali
 sed myeloid signalling environments that mirror the spatial compartmentali
 zation of malignant cells. Our findings define a stereotyped trajectory of
  cancer cells in glioblastoma and unify glioblastoma tumour heterogeneity

DESCRIPTION:Mapping the rules of glioblastoma using spatial genomics\n\nTh
 e malignant brain tumour glioblastoma multiforme (GBM) displays significan
 t levels of cellular plasticity. It is often assumed that the tumour micro
 environment (TME) regulates GBM cell states\, but we know little about the
  spatial organisation of GBM tumours and TME-derived signals driving malig
 nant cell state transitions. Here\, I will present an integrated single ce
 ll and spatial multi-omic approach to resolve the tissue architecture of G
 BM. First\, I will present cell2location\, a new computational tool that c
 an map fine-grained cell types in spatial transcriptomics data. Second\, I
  will present GBM-space\, a new collaborative effort to discover TME-GBM i
 nteractions using multi-modal genomics. Using joint single cell transcript
 omic and epigenomic profiling\, we define recurrent malignant cell states 
 and expand the description of the TME cells. Using Visium spatial RNA-sequ
 encing\, we profile distinct tumour sites and observe significant regional
  heterogeneity of the GBM microenvironment. Finally\, we integrate single 
 cell and spatial transcriptomics using cell2location and discover that mal
 ignant cell states regionally segregate in GBM and associate with distinct
  TMEs. Our efforts reveal the novel spatial organisation of GBM tumours an
 d identify putative TME signals that regulate malignant cell states.\nSpea
