DESCRIPTION:Leukocytes interact physically with other cells using cell sur
 face receptors. The largest group of such receptors are non-catalytic tyro
 sine phosphorylated receptors (NTRs)\, also called immunoreceptors. Althou
 gh NTRs have structurally diverse extracellular domains\, they signal by a
  shared mechanism\, involving phosphorylation of cytoplasmic tyrosine resi
 dues by SRC-family tyrosine kinases. How ligand binding to NTRs induces th
 is phosphorylation\, also called NTR triggering\, remains controversial\, 
 with roles suggested for size-based segregation\, clustering\, and mechani
 cal force. Here we exploit a recently developed cell-surface generic ligan
 d system to explore the ligand requirements for NTR triggering. \n\nWe exa
 mine the effect of varying the ligand’s length\, mobility and valency on
  the activation of representative members of four NTR families: SIRPβ1\, 
 Siglec 14\, NKp44 and TREM-1. Increasing the ligand length impairs activat
 ion via NTRs\, despite enhancing cell-cell conjugation\, while varying lig
 and mobility has little effect on either conjugation or activation. Increa
 sing the valency of the ligand\, while enhancing cell-cell conjugation\, d
 oes not enhance activation at equivalent levels of conjugation. These find
 ings support a role for size-based segregation\, rather than mechanical fo
 rce or clustering\, in NTR triggering\, consistent with the kinetic-segreg
